Optimization of the extraction process of bioactive compounds from Rhamnus alaternus leaves using Box-Behnken experimental design

The Rhamnus alaternus plant is a rich source of excellent bioactive compounds including polyphenols and flavonoids. Yet, the improvement extraction yield total polyphenol content (TPC) flavonoid (TFC) from this has never been inspected in depth. Hence, using 15-run Box-Behnken design, dynamic maceration process was optimized for TPC TFC order to investigate antioxidant capacity various fractions by DPPH assay. Three factors (i.e., ratio solvent-to-solid material plant, stirring speed time), were investigated establishing cost-effective produce extracts phenolic components. With desirability value 0.970, optimal parameters flavonoids as follows: 10:1 mL/g, 518 rpm, 24 h extraction. For such conditions maceration, highest contents experimentally determined at 230.9 mg GAE/g 121.1 QE/g extract, respectively with an IC50 22.5 μg/mL, agreement obtained model values.
